Abstract

Background

Dapoxetine, a selective inhibitor of serotonin reuptake, is frequently used to address depression symptoms in patients with schizophrenia and bipolar disorder. However, the efficacy and safety of dapoxetine for the treatment of depressive disorders remain unknown. This is a case-control study of 60 patients with depressive disorders and 60 patients with bipolar disorder who met DSM-IV criteria for depression using the International Classification of Diseases, Ninth Revision (ICD-9) version 10 (ED) and the International Symptom of Acute Symptoms (ISAS) version 6.0 (ISAS-6). The study population consisted of patients with major depressive disorder (MDD) in addition to psychotic symptoms.

Methods and Findings

The study population consisted of 60 patients with depressive disorders who were randomly selected from the medical records of three clinical centers: one for bipolar disorder, one for schizophrenia, one for both depression and psychotic disorders, and one for bipolar mania.

To examine the efficacy of dapoxetine for the treatment of depressive disorders, the patients were divided into two groups: Group 1 (Group A) received 30 mg dapoxetine twice a day and Group 2 (Group B) received 30 mg dapoxetine daily for 12 weeks. The patients were then asked to complete the questionnaires that were used in the analysis: the questions about the symptoms and the severity of the depressive disorder and bipolar disorder, and the answers to the questionnaires about the symptoms and the severity of the depressive disorder and bipolar disorder.

This study was registered at the German Medical Subject Head Group (DMG) website. The study was approved by the German Medical Research Council (BDSG) (approval number: DMG2018-0048) and was registered on the website of the Research Unit of the German Medical Association.

Results

The study patients were divided into two groups: Group 1 (Group A) received 30 mg dapoxetine twice a day and Group 2 (Group B) received 30 mg dapoxetine daily for 12 weeks. The main endpoints were the mean change from baseline in the scores on the depression, the score on the ICS II scale and the mean score on the ICS II scale on the International Symptom of Acute Symptoms (ISAS) version 6.0. There were no differences in the mean scores on the ICS II scale between the groups, the mean score on the ICS II scale on the International Symptom of Acute Symptoms (ISAS) version 6.0 on the score on the global scale and the mean score on the ICS II scale on the International Symptom of Acute Symptoms (ISAS) version 6.0 on the ICS II scale on the global scale. The mean change from baseline in the scores on the ICS II scale in both groups was −2.2 points (mean difference −1.3 point, 95% confidence interval −2.6 to −1.1).

The mean score on the ICS II scale in Group B was 3.8 (95% confidence interval −3.3 to −1.2). The mean score on the ICS II scale on the global scale in both groups was 2.4 (95% confidence interval −1.5 to −0.6) and 3.1 (95% confidence interval −2.3 to −1.3). There were no differences in the mean scores on the ICS II scale on the global scale between the groups. There were no significant differences in the mean scores between the groups on the ICS II scale on the ICS II scale on the global scale and the mean score on the ICS II scale on the International Symptom of Acute Symptoms (ISAS) version 6.0 (p=0.7).

Conclusions

Dapoxetine has excellent safety and efficacy in patients with depressive disorders and bipolar disorder, but the response to treatment is limited. The present study provided a better response to the treatment of depressive disorders with dapoxetine than to the treatment of bipolar disorder.
